Proteolytic Enzyme Production by Isolated Serratia sp RSPB11: Role of Environmental Parameters
نویسنده
چکیده
An effective proteolytic enzyme producing microbial strain has been isolated from marine habitats and evaluated its extracellular protease production properties with respect to different fermentative physiological parameters. The strain has been identified based on biochemical tests according to Bergey’s Manual of Systematic Bacteriology as Serratia sp and designated as RSPB11. This strain has potential to hydrolyze chitin, gelatin and casein revealing its industrial potential for production of multienzyme complex. Since the isolated strain belongs to Serratia genus, the protease produced by this strain is considered as serralysin and which is not inhibited by PMSF suggesting the enzyme belongs to other than serine type of protease. Further analysis denoted that this enzyme belongs to metalloprotease which is confirmed based on negatively regulation of caseinolytic (proteolytic) activity by EDTA. The maximized enzyme production occurred at medium initially adjusted to pH 7.0 incubated at 33C under aerated environment. Analysis of the pH profile before and after fermentation depicted that irrespective of initial medium pH, it is shifted to pH 9.0 after fermentation suggesting the enzyme produced is alkaline in nature.

Background: Proteolytic enzymes have an important role in variety of physiological and pathological functions. They have been used in therapeutic and pharmaceutical applications. Characterizations of extracellular proteases from various strains of S. marcescens indicate that most strains produce a very similar major metalloprotease. This metalloprotease (serrapeptidase, serrapeptase) is an impo...

This paper describes optimization method that combines the Plackett-Burman design, a factorial design and the response surface method, which were used to optimize the medium for the production of L-asparaginase by Serratia marcescens SB08. Four medium factors, from out of 11 medium factors, were screened by Plackett-Burman design experiments and subsequent optimization process to find out the o...
متن کاملBatch Kinetics and Modeling of Alkaline Protease Production by Isolated Bacillus sp. (RESEARCH NOTE)
The aim of this study was the use of fish waste hydrolysate (FWH) as a substrate for alkaline protease production using isolated Bacillus sp. in a batch system. Then the fermentation kinetics of enzyme production was studied. The results show that with the addition of FWH to the fermentation medium with a final concentration of 4% (optimal concentration), alkaline protease value reached a maxim...
متن کاملEffect of Culture Conditions on the Production of an Extracellular Protease by Bacillus sp. Isolated from Soil Sample of Lavizan Jungle Park
Soil samples of Tehran jungle parks were screened for proteolytic Bacilli. Among eighteen protease producers one of the isolates obtained from Lavizan park, in north east of Tehran, was selected for further experimental studies. This isolate was identified as Bacillus sp. strain CR-179 based on partial sequencing of 16S rRNA. Various nutritional and environmental parameters affected protease pr...
